And I was watching for a particular line I thought I remembered from the movie, and was surprised not to find it. I was sure that in the movie somebody, presumably Cora, said something like "I will have no man if not Uncas!"

That definitely was not one of Cora's lines in the book. Although Uncas was in love with her, she was in love with Duncan. Other than Magua, nobody was bidding for her as a wife, and with her upbringing and personality, she would not have expressed an ultimatum on anything, let alone her own preference of a husband.
You may have run across that line in one of the "B Movie" versions of LOTM or in one of the so-called "sequels" to the book, but James Fenimore Cooper would never have penned a line like that.
